Influence of glucose feeding on the ligninolytic enzyme production of the white-rot fungus Phanerochaete

Abstract: The present work studied the influence of glucose feeding on the ligninolytic enzyme production of in a nitrogen-limited (C/N ratio is 56/8.8 mmol/L) medium. Several sets of shaking flask experiments were conducted. The results showed that 2 g/L glucose feeding on the first day of the culture (24 h after the inoculation) stimulated both fungal biomass growth and enzyme production. The manganese peroxidase (MnP) activity was 2.5 times greater than that produced in cultures without glucose feeding. Furthermore, the glucose feeding mode in fed-batch culture was also investigated. Compared to cultures with glucose feeding every 48 h, cultures with glucose feeding of 1.5 g/L (final concentration) every 24 h produced more enzymes. The peak and total yield of MnP activity were 2.7 and 3 times greater compared to the contrast culture, respectively, and the enzyme was kept stable for 4 days with an activity of over 200 U/L.

Lightweight design of an electric bus body structure with analytical target cascading

Abstract: Lightweight designs of new-energy vehicles can reduce energy consumption, thereby improving driving mileage. In this study, a lightweight design of a newly developed multi-material electric bus body structure is examined in combination with analytical target cascading (ATC). By proposing an ATC-based two-level optimization strategy, the original lightweight design problem is decomposed into the system level and three subsystem levels. The system-level optimization model is related to mass minimization with all the structural modal frequency constraints, while each subsystem-level optimization model is related to the sub-structural performance objective with sub-structure mass constraints. To enhance the interaction between two-level systems, each subsystem-level objective is reformulated as a penalty-based function coordinated with the system-level objective. To guarantee the accuracy of the model-based analysis, a finite element model is validated through experimental modal test. A sequential quadratic programming algorithm is used to address the defined optimization problem for effective convergence. Compared with the initial design, the total mass is reduced by 49 kg, and the torsional stiffness is increased by 17.5%. In addition, the obtained design is also validated through strength analysis.

Abstract:

Pueraria mirifica Airy Shaw et Suvatabandhu is a medicinal plant endemic to Thailand. It has been used in Thai folklore medicine for its rejuvenating qualities in aged women and men for nearly one hundred years. Indeed, it has been claimed that P. mirifica contains active phytoestrogens (plant substances with estrogen-like activity). Using high performance liquid chromatography, at least 17 phytoestrogens, mainly isoflavones, have been isolated. Thus, fairly considerable scientific researches, both in vitro in cell lines and in vivo in various species of animals including humans, have been conducted to date to address its estrogenic activity on the reproductive organs, bones, cardiovascular diseases and other climacteric related symptoms. The antioxidative capacity and antiproliferative effect on tumor cell lines have also been assessed. In general, P. mirifica could be applicable for preventing, or as a therapeutic for, the symptoms related to estrogen deficiency in menopausal women as well as in andropausal men. However, the optimal doses for each desirable effect and the balance to avoid undesired side effects need to be calculated before use.
